体外研究

表油菜素内酯(EBR)是油菜素类固醇,类固醇衍生植物生长调节剂家族的生物活性化合物。将细胞与各种剂量(0-100μM)的表油菜素内酯一起温育24或48小时,并通过MTT测定法测定细胞活力。与LNCaP和DU145前列腺癌细胞中的未处理样品相比,表油菜素内酯以剂量和时间依赖性方式诱导细胞活力丧失。与DU145细胞相比,增加浓度的表油菜素内酯对LNCaP细胞活力丧失更有效,这表明雄激素依赖性细胞对表油菜素内酯比对雄激素非依赖性前列腺癌细胞更敏感。在进一步的实验中,选择25μM的表油菜素内酯,因为其对两种细胞系具有中等的细胞毒性作用。通过在96小时内计数细胞数来检查表油菜素内酯治疗对细胞增殖的影响。在LNCaP中观察到比DU145细胞更高和更早的细胞增殖抑制[1]。

细胞实验

使用LNCaP(CRL-1740)和DU145(CRL-2698)人前列腺癌细胞系。使用PNT1a正常前列腺上皮细胞系。细胞在RPMI 1640培养基中生长,补充有10%胎牛血清,10,000U青霉素/ mL和10mg链霉素/ mL。将细胞在37℃下在潮湿的5%CO 2培养箱(HERA细胞150)中培养。将细胞接种过夜,然后用所需的药物浓度处理。将细胞以每孔1×104密度接种于96孔板中,并暴露于各种浓度的表油菜素内酯24和48小时。向每个孔中加入10μLMTT染料(5mg / mL),并将细胞在37℃下保持4小时。将得到的甲crystals晶体溶解在200μLDMSO中。用分光光度法在570nm处读取溶解的甲amount的密度[1]。
