体内研究 |
在给予LPS之前或之后用羟钴胺素治疗减弱LPS诱导的低血压和血浆RNI的增加并且增强LPS诱导的RNI的尿排泄。在给予LPS(16mg / kg ip)前30分钟给予Swiss-Webster小鼠的羟钴胺素(20mg / kg ip)使LPS的24小时死亡率从80%降低至50%,36小时和96小时死亡率从100降低至100%。至60%(羟钴胺素)[1]。 Hydroxocobalamin被广泛使用,并且似乎是怀疑急性氰化物中毒时的解毒剂。在接受羟钴胺素和高压氧治疗的大鼠中,呼吸改善和紫绀消失,随后平均动脉血压稳定。这些研究结果表明,羟钴胺素和高压氧治疗联合使用对氰化物中毒过程中的脑代谢具有有益和持久的作用[2]。反复给予羟钴胺素没有明显的膈肌和心肌毒性,因此仍然是急性氰化物中毒的安全解毒剂[3]。当NaSH给药后给予羟钴胺素(0.25mmol / kg)时,超过60%的35mg / kg(0.63mmol / kg)NaSH(LD90)小鼠存活(24小时),而小于15%的小鼠没有羟钴胺素而存活。 Hydroxocobalamin(50-100μM)或钴(50-100μM)也可预防由NaSH(500μM)诱导的肝细胞细胞毒性。此外,在NaSH后60分钟加入羟钴胺素仍显示出一定的保护作用[4]。 |

动物实验 |
大鼠:在施用LPS(0.8mg / kg iv)之前30分钟,用无菌PBS(0.1mL / kg静脉内)预处理大鼠。给予LPS后15分钟,当125-介导的血压降低最大时,给予大鼠PBS(0.1mI / kg静脉内)或羟钴胺素(20-30mg / kg静脉内)。测量血压,心率和RNI [1]。小鼠:在标准芯片垫料上进行实验之前,允许25-30g体重的成年雄性CD1小鼠适应环境至少7天。所有动物随意喂食并且在实验前不禁食。用单次注射的每重量比0.1mL / 25g体积处理小鼠。治疗后24小时记录动物的存活[4]。 |
