1. 肝素最初从肝脏中提出，现多由猪、羊、牛等动物的肠粘膜中提取，从肺脏，肝脏出可提取。取新鲜肠粘膜投入反应锅内，按3%加入氯化钠，用40%的氢氧化钠调节pH至9.5。在半小时内逐步升温至60-65℃，继续保温搅拌2h，然后升温至95℃，维持10min，冷却至50℃以下过滤。将强碱性季铵盐阴离子树脂加入滤液中，搅拌吸附8小时，静置过液。次日，虹吸去上层液，树脂用水冲洗至上清液澄清，滤干，用2倍量1.4M氯化钠搅拌2小时，滤干，树脂再用不着倍量1.4M氯化钠洗涤1次。然后，树脂用3M氯化钠洗脱两次，滤干，将滤液合并，加入等量的95%乙醇沉淀过液，收集沉淀，用丙酮脱水干燥，得粗制品。粗制品溶于1%氯化钠中，用盐酸调节pHGCF 1.7-1.8，过滤。用5N氢氧化钠调节pH至11，加30%浓度过氧化氢脱色，过滤。滤液用盐酸调节pH至6，加入等量95%乙醇沉淀。沉啶物用丙酮洗涤，干燥，即为肝素。
