西林醇(0.1-10μM;96小时)单个混合淋巴细胞反应和cona诱导的T细胞增殖呈剂量依赖性。西林醇(10μM)不影响T淋巴细胞的活力。cirsilineol(10μM)对T细胞增殖的抑制不是由于细胞毒性作用[1]。西林醇(1-10μM;预处理3小时)完全抑制IFN-γ诱导的STAT1和JAK2活化的Tyr701磷酸化[1]。细胞增殖试验[1]细胞系:脾细胞浓度:0.1,1,10μM培养时间:96小时结果:抑制单个混合淋巴细胞反应和刀豆球蛋白A(ConA;5μg/mL;72小时)以剂量依赖性方式诱导T细胞增殖。抑制OVA323-339特异性CD4+T细胞增殖。westernblot分析[1]细胞系:脾CD4+T细胞浓度:1,5,10μM孵育时间:预处理3小时结果:完全抑制IFN-γ诱导(25ng;30分钟)STAT1和JAK2激活的Tyr701磷酸化。抑制IFN-γ诱导(25ng;12小时)Th1特异性转录因子T-bet。 |
体内研究 |
西西林醇(3、10和30 mg/kg)通过抑制CD4+T细胞中的IFN-γ/STAT1/T-bet信号,显著改善TNBS诱导的Th1介导的结肠炎。动物模型:8-10周龄雌性C57BL/6、BALB/c和DO11.10转基因小鼠,TNBS(10mg;100μL)[1]剂量:3,10,30mg/kg给药:IP;每日的11天试验结果:对小鼠体重和存活率有显著改善作用。显著减少炎症浸润,恢复破坏性粘膜结构,减轻水肿。 |
